I just added a heater fuel pick up to my 1995 Diesel E350's fuel tank. I plan on adding an Espar or chinese knock off in the future.
I'm not certain about 2006 specifically, but you should have an in-tank pump, a 3/8" engine supply, a 5/16" return, and some wiring.
I've read where some people tap into the 5/16" return line, adding a tee, using it both as a return, and a supply for their heater.
I didn't want to risk upsetting my Diesel's older fuel injection system, so I opted for adding a dedicated draw straw and port to the fuel pump/fuel sender access plate. I used 3/16 steel brake line tube and flare bulkhead fitting. Easy for me to do now, on the bench, because I have both fuel tanks out at the moment.
